733: SPIDAR: Decision-Ready In Vivo Multi-Assay Platform For Analgesic Fingerprints And CNS Tolerability
Topic: Basic Science
Subtopic: Pain category (ICD-11), Neuropathic pain , , , , , , , , , , Drug development
Background and Aims: Early discovery programs in pain require rapid strategies to de‑risk efficacy and tolerability before committing to fully powered, single‑model studies. SPIDAR (Screening Paradigm to Identify Drug Analgesic Response) was developed as a streamlined in vivo screening platform delivering a decision‑ready “analgesic fingerprint” to support early GO/NO‑GO decisions.
Methods: SPIDAR combines 7 validated pain assays run in parallel in mice across key pain dimensions: acute (cold plate; von Frey in naïve animals) and tonic (formalin early/late phases) pain, osteoarthritis ( MIA - weight bearing), neuropathic (SNI - von Frey; oxaliplatin‑induced hypersensitivity - von Frey), and post‑operative pain (Brennan incision - von Frey). In parallel, tolerability is assessed using a Functional Observational Battery (FOB) and LABORAS (Laboratory Animal Behavior Observation Registration and Analysis System) for automated activity monitoring. A screening design is used (typically n=6 per model/test, single dose, single time point), with results expressed as a percentage of activity against the vehicle and summarized visually as radar‑style fingerprints.
Results: SPIDAR was benchmarked and validated using clinically established reference compounds across multiple classes (opioids, NSAIDs, anticonvulsants, antidepressants), including morphine (10 mg/kg, s.c.), celecoxib (60 mg/kg, p.o.), pregabalin (30 mg/kg, p.o.) and duloxetine (30 mg/kg, p.o.), generating compound‑specific analgesic fingerprints alongside integrated FOB/LABORAS tolerability profiles.
Conclusions: SPIDAR provides a rapid and cost‑efficient in vivo screening approach that integrates multi‑dimensional analgesic profiling with early CNS/behavioral tolerability assessment, enabling earlier, better‑informed GO/NO‑GO decisions while supporting the 3Rs principles through reduced animal use and optimized experimental design.
